J2抗雙鏈RNA IgG2a單抗是雙鏈RNA檢測的金標準。起初只用于植物病毒檢測研究,但從2006年Weber et al,發(fā)表文章表明:在檢測的感染細胞內所有的正鏈RNA病毒均產生大量雙鏈RNA,隨后該抗體被應用于廣泛的檢測系統(tǒng),發(fā)表文章超過200余篇。
J2抗體用于檢測病毒dsRNA 中間體,例如:丙型肝炎病毒、登革熱病毒、小鼠腦脊髓炎病毒、鼻病毒、基孔肯雅病毒、狂犬病病毒、脊髓灰質炎病毒、豬瘟病毒、雀麥草花葉病毒檢測樣本可以是細胞培養(yǎng)物或石蠟包埋的組織。
J2抗體被廣泛應用于電鏡學、免疫熒光顯微技術、免疫組化及各種免疫捕捉實驗,如:斑點免疫印跡、ELISA。J2抗體是用于區(qū)分被檢樣本是病毒或細菌的有效工具((Richardson et al., 2010))。近,J2用于制備mRNA過程中dsRNA的監(jiān)測,因此該抗體基因治療中,有潛在的應用價值。

K1單克隆抗體,IgG2a亞型,能夠識別dsRNA(雙鏈RNA)。K1抗體可用于檢測細胞和組織中的dsRNA.經證明,K1抗體是J2很好的替代品。在一些特殊的實驗中,用K1代替J2抗體,可以明顯解決交叉反應或者是高背景。
K1抗體可以檢測病毒dsRNA中間體,例如肝炎病毒、小鼠腦脊髓病毒、乙型腦炎病毒等。K1抗體還可以檢測培養(yǎng)細胞以及石蠟包埋組織中的dsRNA。
如果要檢測Poly I:C(多聚肌苷酸),我們強烈推薦使用K1抗體而非J2抗體,因為K1與合成的多核糖核苷酸有很強的親和力。
K1抗體被廣泛應用于免疫熒光、流式細胞術(FACS)和免疫捕捉檢測,例如斑點免疫印跡及ELISA實驗。

K2單克隆抗體,lgM亞型,是抗dsRNA的單克隆抗體。
K2抗體主要用于一些特殊的檢測。K2抗體主要用于ELISA和夾心法ELISA實驗,一般來說,K2抗體只能用于檢測IgG(IgG2a)亞型抗體不適合的雙鏈RNA(dsRNA)。
K2抗體需干冰運輸,因為高濃度的K2抗體容易發(fā)生凝聚,因為不容易做成凍干粉,不能常溫運輸。(J2和K1可常溫運輸。)

1)J2、K1單克隆抗體由于凍干過程中,稀釋的抗體可能含有少量的變性蛋白聚合物,這些蛋白可能會對免疫組化實驗產生高背景。因此,我們推薦在使用前,抗體重懸后,需離心,使用其上清。
2)J2、K1可常溫運輸,稀釋后請分裝,-20 ℃或-70℃保存,盡量避免反復凍融。
3)K2需干冰運輸,到貨后請分裝,-70℃保存。
